Management highlights
Kymera Therapeutics was founded 9 years ago with the goal of building a pipeline using targeted protein degradation. The company focuses on immunology due to unmet need in accessing effective therapies. Key pillars include clear vision, deep development capabilities, $775 million cash as of 2025Q1, and bringing 5 new molecules to clinic since 2020. Discussed STAT6 program with KT621, upcoming data readouts, IRAF5 program details, and decision to pause TIC2 program to extend cash runway and focus resources on other programs. Jared provided an update on STAT6 program with KT621, including ongoing and planned clinical trials. Veronica introduced IRAF5 program, detailing its potential in treating autoimmune diseases and preclinical data of KT579.
Guidance
Upcoming milestones include STAT6 phase 1a healthy volunteer data in June, phase 1b AD data in Q4, phase 2b studies for STAT6 in AD and asthma starting in Q4 2025 and Q1 2026. IRAF5 program expected to start phase 1 in early 2026. Cash runway extended to first half of 2028.
Risks
Market volatility, competitive landscape, uncertainties in clinical development and regulatory approvals.
Q&A highlights
Q: Can you talk a bit about your confidence in targeting STAT6 in AD and of showing an effect in the four-week study?
A: The underpinning is the data from dupilumab, which blocks IL4 and 13 and shows effects in four weeks. We expect robust data in biomarkers and clinical endpoints given preclinical data showing dupilumab-like activity.
Q: Maybe just one question around the decision not to advance the TIC2 program. You obviously talked about the decision in the context of being capital conscious and the macro backdrop. But it appears you've swapped TIC2 for perhaps IRAF5. So is there anything there in terms of the molecule's profile or just the evolving competitive landscape that influenced your decision?
A: Decision is around being capital conscious and prioritizing resources. TIC2 space has competitive intensity, while IRAF5 is a different program with less competitive intensity and potential for best-in-class in wider diseases.
Q: Just on IRAF5, can you talk a little bit about the relative infection risk given the broader or more pan pro-inflammatory cytokine suppression profile here?
A: IRAF5 is selectively expressed in key immune cells and specifically activated, so it won't cause broad immunosuppression. It leaves IRF3 and 7 intact, which are important for immune surveillance, reducing infection risk compared to broad immunosuppressants.
